Imaging Tumor Acidity: pH-Low Insertion Peptide Probe for Optoacoustic Tomography.
Optoacoustic tomography has been used for the detection of pancreatic ductal adenocarcinoma targeted by pH-low insertion peptide (pHLIP) conjugated to near-infrared fluorescent dye. It was proved that tumor targeting is pH dependent. The approach could have major implication for detection and monitoring of pancreatic and other cancers.

BACKGROUND pH-low insertion peptides (pHLIP) can serve as a targeting moiety that enables pH-sensitive probes to detect solid tumors. Using these probes in conjunction with multispectral optoacoustic tomography (MSOT) is a promising approach to improve imaging for pancreatic cancer. METHODS A pH-sensitive pHLIP (V7) was conjugated to 750 NIR fluorescent dye and evaluated as a targeted probe f...
متن کاملFamily of pH (low) insertion peptides for tumor targeting.
Cancer is a complex disease with a range of genetic and biochemical markers within and among tumors, but a general tumor characteristic is extracellular acidity, which is associated with tumor growth and development. Acidosis could be a universal marker for cancer imaging and the delivery of therapeutic molecules, but its promise as a cancer biomarker has not been fully realized in the clinic. ...
متن کاملA Quick Responsive Fluorogenic pH Probe for Ovarian Tumor Imaging
A novel cell-permeable compound, CypH-1, that is non-fluorescent at neutral pH, but fluoresces under mildly acidic conditions with a near infrared maximum emission wavelength was designed for the detection of tumors in the clinical setting. The potential of CypH-1 in ovarian cancer imaging was demonstrated using a murine model. The intraperitoneally administered CypH-1 results in a robust fluor...

The recurrence of certain cancers remains quite high due to either incomplete surgical removal of the primary tumor or the presence of small metastases that are invisible to the surgeon. Near infrared (NIR) fluorescence imaging might improve surgical outcomes by providing sensitive, specific, and real-time visualization of normal and diseased tissues if agents can be found that discriminate bet...
